Procedure OF - Total Count Of Sperm In Lab

Total Count Of Sperm
     Normal Range Of Sperm Count is 40-100 million/ml. It is done manually by improved neubour counting chamber.

Metarial:-
  • Microscope
  • Clear & Dry Counter.
  • WBC Pipette.
  • Improved Neubour Counting Chamber.

Reagent:-
  • Serum Dialuting Fluid NaHCO3 ( Sodium Bicarbonate).
  • 35% Formalin.
  • Dialluted Water.
 Procedure:-
  1. Take The Semen Sample in WBC pipette upto 0.5 marked.
  2. Take semen analysis fluid upto 10 marked, then mixed well. here the dialution 1:20. then wait for 3-5 minute for spermatozoa to settle.
  3. Take a cover slip on the neobour counting chamber on this counting area.
  4. Discard few drops of mixed semen from wbc pipette. then take one drop of mixed semen at the area of the cover slip on this chamber.\
  5. Then wait for ( 3 min to settle down of sperm ).
  6. Then examined under microscope at 10 x Objective.
  7. Count the total number of sperm in tow large square. multiply the sperm by 100000 & express the sperm count at million/ml.
Semen analysis Fluid composition;-
    Formalin 1 ml + Sodium Bicarbnate 5 gm + D/W 100 ml
