Procedure: Estimation Of -Serum Creatinine In Laboratory

Method:- Jaffy's Reaction (German Scientist - 1843-1911)

Reagent:-

1. Picric Acid Solution - ( D/W + Picric Acid )- { Sediment must be present }
2. 2NaOH ---( 1 L D/W + 80gm NaOH )

Procedure:- 

• 1500 µl Supernatant portion of  Picr Acid.
•  Add 200 µl Patient Serum.
• Then Centrifuge 5 min at  3000 rpm.
• Then 100 µl NaOH take on a vial/test tube.
• 1000 µl Supernatant Portion of Centrifuge Solution on the vial/test tube.
• Wait For 20 min
• Then Reading On Colorimeter. 

If an  Jaffy's Reaction Positive then test will be done at Kinetic Method.

500 µl R1 Creatinine Reagent  + 500 µl R2 Creatinine Reagent.
                                       +
                      100 µl Patient Serum
                                       +
        Then Reading on analyzer within 30 second .
